Hydrogels are crosslinked polymeric networks, which have the ability to hold water within the spaces available among the polymeric chains. Hydrogels have been gaining significant importance due to their potential uses in cell based therapies, tissue engineering, liquid micro-lenses, cancer therapy, cell carriers and/or entrapment, wound management and drug delivery. They are sensitive to environmental stimulus like aqueous media, pH, temperature, glucose concentration, electric field, light, etc. A number of synthetic hydrogels such as poly (N-isopropylacrylamide) (pNIPAm), polyethylene glycol (PEG), polyvinyl alcohol methylacrylate co-polymers (PVA-MA) and polylactic acid (PLA), as well as some of the natural hydrogels such as Fibrin, Hyaluronic acid (HA), Agarose, Alginate and chitosan are available. Because of the compatibility with living tissues, hydrogels can be used in different medical purposes (for making contact lenses, stents, balloon catheters, artificial muscles, substitutes for arteries and veins, trachea, oviduct, for neutraceutical delivery, regenerative medicine). Self-healing materials are of great importance as they offer longer lifetime, low replacement cost and greater product safety and can be used in any stress-bearing applications. Pioglitazone HCl was used for the management of type-2 diabetes. Here an attempt was made to develop the floating matrix tablets, which was designed in the way that after oral administration the GI resistant time is prolonged and thus give sustained action with increase in bioavailability of the drug. Sustained release tablets were developed by using a combination of hydrophilic polymer (hydroxypropyl methylcellulose) and effervescent substance (sodium bicarbonate and citric acid) in different ratios. The prepared tablets were characterized by drug content, floating property, swelling and in-vitro dissolution test. Drug polymer compatibility studies by FTIR gave conformation about drug purity and showed no interactions between drug and selected polymers. Among all the formulations, the formulation F9 containing drug and polymer in the ratio of 1:3 extended the drug release upto 12 hrs. The drug release of optimized formulation (F9) follows the first order kinetics and found to suitable for once a day tablet for pioglitazone....
The primary aim of this study was to prepare a buccoadhesive tablet of nimodipine by kneading technology, using optimized concentration of polymer to give maximum retention effect with good release profile. Nimodipine is a 1,4-dihydropyridine calcium channel blocker mainly used for prevention of cerebral vasospasm and resultant ischemia. It is having a low oral bioavailability and undergoes hepatic first pass metabolism. Solubility of nimodipine was enhanced by complexing it with β cyclodexrtin. Prepared tablets were subjected to various physicochemical parametes. In this study, buccoadhesive tablet were prepared which having backing layer of magnesium stearate and ethyl cellulose for support. It was concluded on the basis of buccoadhesive strength and in-vitro release kinetic that optimized formulation, F9 gave the best in-vitro release of drug over a period of 6 hrs....
Spansules are one of the most advanced and modified form of drug delivery system. Spansules gives one of the best methods to deliver multiple drugs at one time. Since the spansules have different coating thickness, they release drug at different manner. The entrance of the spansules into the small intestine (where the majority of drug absorption occurs) is usually more uniform than with non-disintegrating sustained-release tablet formulations. In case of controlled release spansules the onset of absorption is less sensitive to stomach emptying and reduces the rate of rise of drug concentration in blood and enhanced bioavailability. Spansules increases the safety margin of high potency drugs and reduces the incidence of both systemic and local side effects in sensitive patients. Spansules reduces the dosing frequency, improved patient compliance and reduce side effects. Spansules provide both controlled and sustained release for single or multiple prescriptions and over the counter medicine. Present review discussed on the spansules as one of the novel dosage form, various pharmaceutical advantages of spansules over conventional dosage forms, different evaluation parameters of spansules and available marketed preparations of spansules. The controlled drug delivery system is become the fastest growing field in pharmaceutical dosage forms and acquiring dominance in market. So many product in market are in available in spansules capsules dosage form.
